1) exotic, dagger, polearm, sword or axe are needed to behead with *regular*
attacks. This is where the beheading skill (and sharp/vorpal flags) come into
play. Double/round swing (and a few others) are considered to be regular
attacks.
2) Assassination requires a dagger or ranged weapon. While you can circle with
a sword, you can't assassinate someone with it.
3) Unarmed attacks can also behead/assassinate, but only with the right skills:
shadowclaw enables unarmed beheads, while lethal hands enables unarmed
assassination.
4) Daggers increase the anatomy damage bonus to circle/backstab (or rather, it
is halved with other weapons).
